Large Health-Record Study Finds Link Between Glucosamine Use and Faster Progression from MCI to Dementia

New observational analysis of medical records has found that people taking glucosamine, a widely used joint supplement, were more likely to see mild cognitive impairment progress to dementia. The study reports a roughly 25% higher likelihood of progression among glucosamine users compared with non-users, based on large-scale health data. The finding adds to questions about the supplement’s effects on brain aging and cognitive decline.

The research team combined the epidemiological analysis with laboratory experiments that probed possible biological pathways. Those lab results indicated that exposure to glucosamine could enhance an abnormal sugar-tagging process observed in brains affected by Alzheimer’s disease. Investigators say the experimental data provide a plausible mechanism linking the supplement to molecular changes associated with the disease, but they emphasize that these laboratory observations do not by themselves establish harm in people.

Experts involved in the work underline the limits of the available evidence. Because the clinical component was observational, it cannot prove that glucosamine causes acceleration from mild cognitive impairment to dementia — unmeasured factors could contribute to the association. The authors call for randomized clinical trials to test causality and to quantify any potential risk, and they note that further studies should seek to replicate the finding in other populations and assess dose and duration effects.

The report poses a practical challenge for clinicians and people who use over-the-counter supplements for joint health. Until randomized trials are completed, physicians and patients will need to weigh these emerging findings alongside other evidence about benefits and risks. Researchers say the next steps are clear: confirmatory studies, carefully designed trials, and mechanistic work that can determine whether the laboratory signal translates into a clinically meaningful effect. Given the popularity of glucosamine, any confirmed link to cognitive decline would carry significant public-health implications and warrant clear guidance from the medical community.
